Sažetak
Despite the considerable efforts of the last years to increase the understanding of mechanisms and regulation of cellular metabolism the knowhow is still not sufficient to remove taste of applied research from the mammalian cell culture. Mammalian cell culture is known to be a necessary tool for the production of complex proteins as well as for other applications for which microbial cells are not suitable. But the potential of the smallest independent living parts of all living organisms is not exhausted with the expresion of heterologous proteins. The importance of mammalian cell culture for the production of biopharmaceuticals is evident from a several papers related to "biotechnology medicines in development". From the particular interest is aplication of cultured cells as in vitro test systems in study of toxicity, immunology, drug side effects, pharmacokinetic studies, cosmetic compounds or environmental influences. In vitro cultivated mammalian cells require a larger number of specific and much more precise physical and chemical conditions of their micro- and macro- environment compared to microbial cells. In the toxicology most research work is still done with animals, with organs and tissues in situ or with isolated organs ex vivo. Studies with cell cultures can reduce and sometimes replace experiments with animals. The advances in the areas of mammalian cell culture and biotechnology made it possible to develop in vitro protocols for application in toxicity testing. Due to severe environmental demage reprotoxicity and teratotoxicity are becoming other important fields for applied research activities with cell cultures initiated only recently.
