Sažetak
The usefulness of animal cell technology became very important since it was shown that this was the unique way to obtain products with application in areas such as veterinary and human health, either as vaccines, therapeutics or diagnostics. In this work BHK 21 C13 cells growing in suspension were used as a model in production of cells biomass. To obtain high cell density, influence of different amounts of CABRPP as growth promoter were investigated. The basic medium used was Glasgow BHK 21 medium (Gibco) supplemented with foetal calf serum (FCS). Cells in log phase were seeded in T-flasks in appropriate medium supplemented with 5 % FCS in atmosphere of 5 % CO2 and 95 % air at 37 0C. During cell growth the total cell density, viability and growth kinetics were determined. Results obtained with basic medium supplemented with 5 % FCS and initial cell concentration of 1.8 x 10^5 cells/mL were: cell density 11.9 x 10^6 cells/mL, viability was 95 % and tD =15.6 h. After addition of CABRPP, cell density increased on 2.3 x 10^6 cells/mL, viability was 95 % and tD = 12 h Application of CABRPP provides reduced amount of neccesary serum addition and significantly increases a biomass production.
