Sažetak
Apo E is a polypeptide synthesized primary by liver and brain, and less amounts by ovary, adrenal gland, spleen, lung, heart, kidney, muscle and skin. It plays essential role in lipid metabolism ; not only in the uptake and degradation of acyl-glycerol lipoproteins, but also in intracellular cholesterol redistribution. The apo E gene is located in chromosome 19 consisting of four exons and three introns. There are three common alleles (e2, e3 and e4)inhereted in autosomal codominant manner, giving six common genotypes. The molecular basis of apo E polymorphism are cysteine-arginine interchanges (E2:Cys Cys112/Cys158, E3: Cys112/Arg158, E4: Arg112/Arg158). The aim of our study was to investigate the connection of apo E genotype with neurologic disprders. Apo E genotype was determined by the PCR-RFLP method in a group of healthy volunteers (n=30)and 30 patients with extracranial carotid artery stenosis (stenosis more than 50%). All patients were examined with color Doppler flow imaging in order to asses the stenosis degree. The frequency of diagnosed apo E genotypes was as follows: E3/3 76%, E2/3 10% and E3/4 13.3% in control group, and E3/3 43.3%, E2/3 36.6% and E3/4 20% in the patient group. Our preliminary results indicated as association between neurologic disorders and apo E genotypes (more E2/3 and E3/4 genotypes and less E3/3 in comparison with the control group), and importance of further apo E role investigation in the nervous system pathology.
