Sažetak
The induction of micronuclei (MN) in human lymphocytes can be used as a measure for the cytogenetic damage. The aim of this work was to study the induction, frequency, distribution and Ag-NOR activity in micronuclei of medical personell who administered antineoplastic drugs. For the study healthy non-smoking female subjects were chosen. Their average age was 39.5 years, and their average occupationally exposure to antineoplastic drugs was 17.3 years. For the micronucleus assay whole blood samples were cultivated for 72-hour and cytokinesis was blocked with cytochalasin-B. Slides were stained by use of conventional Giemsa staining and silver staining technique. For each staining technique 500 binucleated cells per subject were analysed. Both techniques revealed an increased number of MN (up to 30 MN/500 cells) in all subjects compared to control (5MN/500 cells). In respect to the patterns of nucleolar organizer (NOR) activity in MN we distinguished Ag-NOR+ and Ag-NOR- micronuclei. The higher proportion of Ag-NOR+ MN observed in all subjects indicate the involvement of Ag-NOR chromosomes (D and G groups) in the formation of micronuclei.
