Sažetak
For the evaluation of genotoxic effects of antineoplastic drugs different cytogenetic techniques are used. The aim of this study was the determination of cytogenetic damage in medical personell who handle with antineoplastic drugs. Samples of peripheral blood were taken from healthy nonsmoking female subjects. Their average age was 39.5 years, and their average occupationally exposure to antineoplastic drugs was 17.3 years. The determination of micronuclei (MN) was made in cytokinesis blocked lymphocytes stained with DAPI (4', 6-diamidino-2-phenylindole-dihydrochloride) and Giemsa staining techniques. For each staining technique 500 micronucleated cells per subject were scored. The total number of MN detected by use of both staining techniques was increased in all subjects compared to control (5MN/500 cells). In Giemsa stained cells up to 30 MN/500 cells, and in DAPI stained cells up to 29 MN/500 cells were observed. DAPI staining technique gave more informations about the origin of MN and allowed us to distinguish between MN+ (MN with one or more bright fluorescing DAPI-dots) and MN- (MN without bright fluorescing DAPI-dots).
