Sažetak
A new, rapid and sensitive technique for visualising and measuring DNA damage and repair in individual cells is the Comet assay or single cell gel (SCG) assay. This technique detects DNA strand breaks, alkali-labile damage, incomplete excision repair sites and DNA:DNA crosslinking at the level of individual cells. Since the development of the comet assay, the damage and repair of single-strand DNA breaks can be rapidly and accurately determined using an epifluorescence microscope. This technique has been applied to a number of plant tissues and animal tissues and cells as well in human biomonitoring.In recent experiment we show that the comet assay is very sensitive in assessment of DNA damaging effects of antineoplastic drugs in subjects involved in their preparing and application. In conclusion, Comet assay might be helpful in analysing of genotoxic agents and applied as a rapid additional test in human biomonitoring.
