Sažetak
The aim of the present study was to investigate 4 Short Tandem Repeat (STR) loci (LEI07, LEI08, LEI22, and LEI32}; ; in the population of the Croatian shepherd. The loci were selected on the basis of literature data. The sample was comprised of 45 unrelated dogs from Slavonski Brod, Županja and Đakovo. All dogs had phenotypic characteristics of Croatian shepherd. After DNA isolation, samples were amplified in 4 separate PCR reaction and run on a 6% polyacrylamide gel in an ALFexpress sequencer (Pharmacia Biotech, Uppsala, Sweden). The results were analysed using AlleleLocator program (Pharmacia Biotech, Uppsala, Sweden). At 4 tested STR loci, the number of heterozygotes was in range of 54.3% (LEI32}; ; and 77.8% (LEI08}; ; . Nine different alleles were observed at LEI07 locus, 8 alleles were found at LEI08 locus, while 6 alleles were observed at the LEI22 and LEI32 loci each. We compared our results with the data from literature for other Croatian canine species (Istrian hound, Dalmatian dog). Comparison demonstrated that some alleles specific for Croatian shepherd were not observed in the other two species and vice versa. This study is the beginning of the creation of a database containing information about the polymorphism of the STR loci in the population of the Croatian shepherd.
