Sažetak
With aim to test appropriation cpDNA analyses for intra and interspecific phylogenetic study of genus Hypericum, preliminary investigations of cpDNA polymorphism were performed in 5 species: H. perforatum subsp. typicum Beck, Hypericum perforatum subsp. veronense (Schrank) Beck, Hypericum perforatum subsp. angustifolium DC., Hypericum maculatum Crantz, Hypericum hirsutum L., Hypericum androsaemum L. and Hypericum calycinum L. comparing restriction patterns of cpDNA fragment amplified by PCR with two pairs of universal chloroplast primers. Considerable degree of interspecific polymorphism was detected in both restriction profiles while intraspecific pattern was identical even between H. perforatum subspecies. This results improved some degree of rearrangements in cpDNA molecule during species separation which is comparable with subgenes section aggregation of species performed by classical morphology based systematics. Also highly conservation of cpDNA on lower phylogeneticaly level excluded this method in intraspecific taxonomical investigation in genus Hypericum.RAPD analysis of genome as well as phytochemical investigation of Hypericum perforatum subspecies are in process.
