Sažetak
Adjuvant properties of PGM have clearly been demonstrated so far, but in comparison to more potent commercial adjuvant its effectiveness has been low. We hypothesized that the reason for this is the short life of PGM in the mammalian organism due to its fast excretion and degradation. Our efforts have been directed recently to a design of novel adjuvant formulation where PGM is incorporated into an oil-based adjuvant in order to prolong its life, and hopefully its effectiveness. Incomplete Seppic Adjuvant 720 (ISA 720, Seppic, France) is a representative of novel generation of oil-based adjuvants. When mixed with water-phase containing antigen of interest it forms stable water-in-oil emulsion that is currently under Phase I and Phase II clinical trials. The aim of our study was to incorporate PGM into an ISA 720 w/o emulsion together with an antigen (human IgG) and to see whether such adjuvant formulation would induce higher quantity of antigen-specific antibodies upon immunization of experimental animals. Results indicated that the immunisation of rabbits with the antigen (hIgG) incorporated into novel ISA 720 + PGM adjuvant formulation induced statistically significantly higher quantities of antigen-specific antibodies, measured by ELISA, in comparation to both, group immunised with the antigen only and what is more important, with the antigen in ISA 720 w/o emulsion. Higher quantities were detected after each of all three immunisations of rabbits. Also, the novel adjuvant formulation induced higher quantities of specific antibodies than Complete Freundşs Adjuvant immunised group developed, suggesting that ISA 720 + PGM could be a very potent novel adjuvant formulation. We have also started the experiment using ovalbumin (OVA) incorporated with PGM into an ISA 720 w/o emulsion. Preliminary results indicate that in rabbits such combination induces very high levels of specific antibodies.
