Sažetak
In this study we have used the newly introduced MHC tetramer technology to evaluate specific T cell response in influenza infected patients. Analysis of the HLA-A*02 allele frequencies was performed in the group of 68 HLA-A2 positive healthy individuals. High resolution HLA-A*02 typing revealed five different HLA-A*02 subtypes (*0201, *0203, *0205, *0206, *0217) with the prevalence of A*0201 allele (92%). Group of 29 patients with influenza infection were recruited from Institute of Immunology for HLA class I typing. Thirteen patients were positive for HLA-A2 antigen and the A*02 high resolution typing revealed the presence of HLA*0201 subtype in 11 patients while the remaining two patients carried A*0205 and A*0203, each. Using the custom designated HLA-A*0201-restricted influenza matrix protein tetramer the enumeration of CD8+ influenza specific T cells was achieved in A*0201 healthy individuals and in 11 A*0201 influenza infected patients. The results showed the presence of influenza specific T cells only in HLA-A*0201 patients who were in acute phase of infection, while these cells were not present in peripheral blood of HLA-A*0201 positive individuals without influenza infection.
