Sažetak
One of the most relevant topics in today’s environmental analytical chemistry is water quality. In order to characterize water quality, concentrations of organic pollutants have to be determined. Recent studies have shown that a multitude of drugs are present in aquatic systems, what include also antibiotics. Until recently exposure of antibiotics to environment has not been considered a big concern because they were thought to be safe. Nowadays, however, they are referred to as the emerging contaminants. For majority of the emerging contaminants, data about their occurrence, related risk assessment and ecotoxycological data are not available. The interest in the analysis of antibiotic residues in the environment arises from the fact that they are suspected of being responsible for the appearance of bacterial strains that are resistant to antibiotics which are important drugs for the treatment of many serious infections. [1-3] This work focuses on the application of SPE-HPLC analysis of important veterinary antibiotics from different classes in highly complex wastewater matrix. The antibiotics investigated included three sulfonamides (sulfaguanidine, sulfadiazine and sulfamethazine), a sulfonamide synergist from diaminopyrimidine class (trimethoprim), a tetracycline (oxytetracycline), three fluoroquinolones (ciprofloxacin, enrofloxacin and norfloxacin) and a β-lactame (penicillin G/procaine). The method involves pre-concentration and clean-up by solid-phase extraction (SPE) using Oasis HLB (Waters) and StrataX (Phenomenex) extraction cartridges. The antibiotics were extracted from wastewater samples (100 mL) with methanol (pH=4.0). Final analysis of selected antibiotic compounds was carried out by high- performance liquid chromatography (HPLC) with gradient elution of mobile phase (0.1% formic acid in acetonitrile and 0.1% formic acid in water) on C18 and CN modified column and detected by mass spectrometry (MS) via an electrospray ionization interface (ESI) in positive mode. The validation method yielded good results and included the range, linearity, recovery, limit of detection (LOD) and limit of quantification (LOQ) values.
