Sažetak
Using matrix-assisted laser desorption ionization time-of-flight imaging mass spectrometry (MALDI-TOF-MSI), the cerebellum of KO and WT (wild-type) 3-, 9- and 20- month-old mice (4 animals per group) was analyzed. A 9-aminoacridine matrix was applied to 16 μm thick fresh frozen tissue sections mounted on indium tin oxide coated slides. Further analysis was performed in negative and positive mode in the range 400 - 900 m/z. Data were analyzed using the software package IMAGEREVEAL (Shimadzu Inc.). Statistically significant m/z values were identified using Human Metabolome Database. Analysis was focused on cerebellar white matter as the pathophysiological substrate of the phenotype. The greatest differences were observed in negative mode between 20- month-old WT and KO mice. All statistically significant m/z values favored WT mice. Among the uniquely identified compounds were: guanosine nucleotides, cytidine triphosphate (CTP), N-acetylated amino sugars and fumarycarnitine.
