Sažetak
The neocortical six-layer lamination is established by a series of prenatal histogenetic events via transient fetal lamination. Deep layer projection neurons (DPN) belong to the early-born cell population, involved in the correct formation of the transient laminar structure that is a prerequisite for establishment of the regional cortical architecture. However, the laminar dynamics of the DPN transcriptional factor markers is still poorly understand. Here, our objective was to analyze early regional differences that appear at 8 postconceptional weeks (PCW) during the initial formation of the cortical plate (CP) phase of the developing human frontal lobe.To visualize expression pattern of DPN markers during the initial CP phase we used immunofluorescence doublelabeling method on postmortem prenatal human brain tissue. FFPE sections of postmortem brain tissue, that are part of Zagreb Neuroembryological Collection were stained using immunofluorescence (IF) protocol and images were acquired using inverted confocal microscope Olympus FV3000. DPN markers expression pattern shows regional lateral to medio-dorsal differences in the prospective frontal lobe at 8 PCW. Importantly, initial CP is present in the lateral portion of the developing frontal cortex, that is clearly visualized by DPN markers. In addition, pioneering cortical plate (pCP) is detected in the dorsal portion, while the sparsed cells with simple organization represent late preplate (PPL), is detected in the medial portion of the developing frontal cortex. Differential expression pattern of DPN markers serve as a good indicator of the CP development, as well as its cytoarchitectonic composition. Our data confirm vast diversity of the molecular markers in the developing frontal neocortical wall and future studies are need to further elaborate these early neurogenetic events.
