Sažetak
Repetitive mild traumatic brain injury (rmTBI) has been connected with cognitive and behavioral deficits as well as progressive neurodegenerative diseases, in some of which TAR DNA-binding protein 43 (TDP-43) dysregulation has been identified. With the aim to research different therapeutic approaches for the mentioned disturbances and diseases in the future, in this study we investigated the effects of repeated concussions on the learning ability and the hippocampal expression of some markers of brain inflammation and stress in mice overexpressing mutant TDP-43, during the first week following rmTBI. Animals were subjected to mild traumas twice daily, 6 hours apart, during 5 days. One day after the final injury/sham procedure, mice were tested on the passive avoidance apparatus. Animals were sacrificed at 1, 3 or 7 days after the last trauma/sham procedure and their hippocampi were prepared for the analyses. We found no significant effect of rmTBI on the learning performance of TDP-43 transgenic mice. In the injured mice sacrificed at day 1 following rmTBI, we detected increased hippocampal expression of COX-2 while the levels of iNOS, HSP70 and cytoplasmic NFκB expressions were unaffected.
