Sažetak
In forensic pathology, the vitreous humour of the eye is a unique biological substrate useful for assessing the post-mortem interval (PMI). Due to its anatomic location, the eye is protected from the influence of external factors and bacterial contamination, which is why the final phase of post-mortem degradation by decay is delayed for a relatively long time. Research to date has mainly been conducted in humans, but it does not differ significantly from research conducted in some animals such as red deer, horses, dogs, donkeys and domestic pigs. The preliminary research was conducted on a total of four wild boars. Immediately after they were shot, both eyes were removed, one eye for histological testing and the other to determine the biochemical parameters. The eyes intended for biochemical testing were stored at +4oC and the vitreous humour was sampled at intervals of 0, 6, 12, 24, 48, 72 and 120 hours, and stored at -20 oC until analysis. Before the analysis, the samples were thawed, centrifuged and analysed using an Abbott Architect c4000 automatic biochemical analyser (Abbott Park, Illinois, USA). The eyes for histological testing were fixed in the same time intervals using Davidson’s fixative, and after the standard processing procedure for histological samples, they were examined under a light microscope Panthera U (Motic, Xiamen, China). Statistical analysis of the correlation between the biochemical parameters and PMI was conducted using the Statistica computer program (Statistica 10, StatSoft Inc.), using simple regression analysis. The statistical hypotheses were tested to a level of significance of p<0.05. Histological tests were based on a subjective assessment of the changes to the structure of the eye. The correlation between the concentrations of biochemical parameters in relation to the PMI were shown to be statistically significant. The concentrations of sodium, chlorine and glucose decreased, whilst the concentrations of potassium increased in relation to the PMI. Histological tests of the eye showed the most important changes in the structure of the retina. With the advance of the PMI, there was an evident rise in the number of cells with pyknotic nuclei and loss of cell structures from the ganglion cell layer to the outer nuclear layer of the retina, an increase in the degree of vacuolation of the cytoplasm of the ganglion cells and the nerve fibre layers, homogenization and compaction of the outer and inner plexiform layers, and the loss of cone and rod cell structures. After 120 hours, subtle changes were visible in the structure of the sclera and cornea. These changes in the concentration of biochemical parameters are related to the halting of cell metabolism after death and the decay of cell structures. Since a very small number of cells are found in the vitreous humour, changes in the concentrations of the biochemical parameters are directly linked with the disintegration of the cell structures of the retina. Apart from the fact that the results of the research indicate great potential for assessment of PMI, they will also represent a basis for further planned research which will include a larger number of animals, and enable comparison with the results of research in the very closely related domestic pigs.
