Sažetak
Protists serve numerous ecological functions in aquatic habitats, but they receive less attention than other aquatic organisms (e.g. macroinvertebrates) and their biodiversity is still poorly investigated. Generally, understanding protist diversity is critical to the specific role of freshwater periphyton in effective management and conservation of freshwater river resources. The Krka River (southern Croatia) is a specific karst river, characterized by its geographic and hydrological uniqueness, habitat heterogeneity and the high biodiversity of aquatic organisms, especially protists. The main aim of the study was to obtain a detailed overview of protist community structure in the periphyton and to determine the differences in protist diversity along the Krka River. Protist community structure was examined by amplicon sequencing of hypervariable V9 region of the 18S rRNA gene. Diversity was explored using measures of alpha and beta diversity. Alpha diversity showed an increasing trend in the downstream direction of the River. Regarding beta diversity, the strong location effect, or grouping of samples by location, was confirmed by the PERMANOVA permutation test. The combination of alpha and beta diversity can provide deeper insight into protist diversity patterns, but can also indicate potential decline in species diversity, and should therefore be implemented in future management and protection of aquatic karst habitats.
