Sažetak
AIM: Metal parts of orthodontic appliances can easily corrode and release metal ions into saliva. Beside beneficial, metal ions could have negative biological effects to humans, so a question, whether those eluted ions can cause damage to patients, rises. MATERIALS AND METHOD: In this paper, four human cell lines (Hep G2, CAL 27, AGS and Caco2) were used to test cytotoxicity and genotoxicity of metal ions eluted from orthodontic appliances. Chosen cell lines are the type of cells that could be the most affected in vivo by those metal ions. Concentrated artificial saliva solutions (Toni-Zucchi artificial saliva, pH 4.8), in which parts of orthodontic appliances (brackets, arches, ligatures, rings and all parts together) were immersed for some time (3, 7 and 14 days) were used as test samples. The cytotoxic effect of all samples at four different concentrations (0.1 ; 0.5 ; 1.0 and 2.0) was examined on tested cell lines, and the Neutral Red method was used. Genotoxicity was measured with Comet assay. JASP statistics program was use for data analysis. RESULTS: The results show that the most sensitive cells of the oral cavity epithelium, CAL 27, and liver cancer cells - Hep G2 were. The cytotoxic effect was caused by metal ions in solutions (especially Ni2+, Cr3+ and Co2+). Cytotoxic effect increases with increasing elution time due to the increased concentrations of metal ions in these solutions. Gastric carcinoma (AGS) and colon cancer cells (Caco2) were resistant to the cytotoxic action of eluted metals. Statistically significant genotoxicity compared to the control show only few tested samples (with arches eluted 3 days on Caco2 and with all parts eluted 3 days on AGS). These samples have an increased concentration of Ni2+, Cr3 and Al3+. Hep G2 cells showed much lower values of tail intensity after treatment with all tested eluates compared to the positive control, indicating activation of protective mechanisms. CONCLUSIONS: Generally, metal ions present in the tested eluates do not cause permanent changes of the genetic material and their application is safe.
