Sažetak
SSB class of bacterial proteins, as well as their eukaryotic RPA analogues, are essential and ubiquitous. They bind single-strand DNA (ssDNA) and regulate/coordinate its metabolism. The prototypic SSB protein, the one of Escherichia coli, is encoded by an ssb gene. Although the ssb gene promoter harbors SOS box, multiple studies during several decades failed to determine the mechanisms of the ssb gene regulation, i.e. whether it is SOS dependent. SOS regulon is comprised of about 40 genes that are induced in stress conditions in a transcriptionally coordinated way. Using quantitative real-time PCR, we determined the ssb gene expression in UV- and γ-irradiated E. coli and revealed that the ssb gene expression is induced in irradiated cells in an SOS-dependent manner. Elucidating regulation of expression of the essential bacterial gene such as the ssb can be very useful in finding ways of combating bacterial infections.
