Sažetak
Ultrasound transvaginal ovum pick-up was performed in 4 Simmental cows suffering from reproductive disorders: cystic ovaries (n=3) and postpartum anoestrus due to endometritis (n=1). Rectal and ultrasound examination were performed during 1 month. Cows were treated with GnRH and PGF2a analogue. After resumption of normal cycle they were synchronized with PGF2a.Transvaginal ultrasound-guided oocyte collection was performed on day 3-4 of the cycle, twice a week for 4 consecutive weeks (OPU-A). After a recovery period of 2 consecutive oestrous cycles cows were stimulated with pFSH. Oocyte retrieval was performed every two weeks: 3-4 OPU sessions per cow (OPU-B). Oocytes were in vitro matured, fertilized and cultured. In non stimulated cows (OPU-A) the recovery rate was 43.6%, 1.9 COCs/OPU. In stimulated cows (OPU-B) recovery rate was 62.7%, 5.9 COCs/OPU. In vitro culture results for OPU-A and OPU-B were 76, 3% and 89% cleavage rate on Day 2, 54, 2% and 41.1% Morula/Blastocysts on Day 7 and 22% and 21.9% Hatched Blastocysts on Day 9 of culture respectively. The average number of embryos per OPU session were 1.84 and 4.87 Morula/Blastocysts and 0.41 and 1.01 Hatched Blastocysts per OPU-A and OPU-B respectively. More embryos were obtained in the FSH stimulated cows. There is no significant difference between problem cows and healthy donors from our previous research which indicated that problem cows could successfully be used for embryo production.
