Sažetak
Proton pump inhibitors, such as lansoprazole (LPZ) are gold standard in the treatment of ulcer disease. However, various studies have shown effectiveness of garlic extracts (GE) in the treatment of ulcer disease. The aim was to determine inhibition of sodium taurocholate (NaT) caused oxidative stress in human gastric cell line (AGS) by treatment of GE and LPZ. Cells were pre- treated with LPZ and increasing concentrations of GE. The ability of GE to protect the AGS cells against the damage induced by NaT was evaluated by glutathione (GSH) and prostaglandin E(2) content by ELISA, AGS proliferation by cell counting and by expression of superoxide dismutase (SOD) and factor kappa B subunit 2 (NFKB2) by RT PCR. Cells pre-treated with LPZ (p<0.001) and GE (p<0.001) showed significant increase in viability, compared to the untreated control. Pretreatment with LPZ and different concentrations of GE were able to increase PGE2 and GSH concentrations in cell culture model of ulcer disease (p<0.001). Positive correlation of SOD, and NFKB (p<0.01) with LPZ and GE pretreatment has been demonstrated. Hence, GE pretreatment showed synergistically gastroprotective effect with LPZ, although, further experiments are needed to fully elucidate protective role of GE in ulcer disease.
