Sažetak
Fatty liver disease is one of the most severe side effects of amiodarone and tamoxifen varying from simple steatosis to end-stage liver disease. In this study, we developed in vitro models of drug- induced fatty liver disease and analyzed the potential protective effect of glucagon-like- peptide-1 receptor agonist (GLP-1 RA). HuH7 cells were incubated with 5 µM to 20 µM of amiodarone, 1 µM to 10 µM of tamoxifen, and 1 nM to 100 nM of GLP-1 RA for various time periods (from 24h to a few days), respectively. Cell viability was measured by MTT assay. Changes in cell shape and the extent of hepatosteatosis were assessed by fluorescent microscopy. A significant decrease in cell viability occurred with tamoxifen administration in a dose-dependent manner after only 24h, whereas amiodarone did not diminish cell viability significantly in such a short period of time. Along with the decrease in cell viability, changes in cells appearance were also observed. Microsteatosis was demonstrated as the predominant form of injury. Co-treatment with GLP-1 RA enhanced cell viability, especially in tamoxifen- treated cells for almost 30% (p<0, 05), and this effect was dose-dependent. Further research is necessary in order to better elucidate the different effects of the abovementioned drugs in hepatic cell cultures.
