Sažetak
Preliminary experiments investigating bud, shoot, and needle generation from shoot-tip explants of 20-day-old and 1-year-old seedlings, and from explants of resting vegetative buds of both juvenile and older P. nigra Arn. and hybrid P. nigrosylvis Vid. trees have been completed. As an axillary-bud induction medium 1/2 MS agarified basal medium supplemented with BA, vitamins and glycine was used. Elongation of induced buds/shoots was obtained on hormone-free medium. Subcultured elongated shoots were individually returned on the induction media to perform the second generation of shoots/buds. This procedure was successful in explants from seedlings and from juvenile trees, three years old or less. Best results were obtained when single shoots were serarated after clumps of shoots were cultured together for some time on hormone-free medium. Besides age of the donor plant, its genotype and the season of donation affected bud regeneration. The most success was reached in early spring. Bud induction from cloned culture-origin shoots were shoving similar season dependence.
