Sažetak
The molecular-biological research has been successfully applied to systematic and phylogeny of plants. Unfortunately, algae have been researched extremely rare in this way. In this research we tried to find out the relevant method for DNA isolation of Chara fragilis Desv. For this purpose modification of Random Amplified Polymorphic DNA (RAPD) technique was used. The isolation of DNA was conducted on the fresh, frozen (-20 oC) and material stored in 95 % ethanol. Isolation from the frozen material and the material that was stored in 95% ethanol did not give satisfactory results. The DNA was successfully isolated, and RAPD technique was efficacy applied on the fresh material. Three of nine tested primers (OPB-11, OPB-12, OPB-18) could be used in prospective systematic and ecological research among the species of Chara genus.
