Sažetak
Introduction: Portable handheld devices used to measure and/or monitor concentrations of different analytes in biological fluids have many advantages. They may be used for medical purposes both in medical facilities or in patient's homes, for educational purposes and in scientific work. Ion-selective electrode (ISE) pocket meters have also become available for measuring concentrations of different ions in various samples, including saliva. As centrifugation is standard procedure prior to performing salivary assays, the aim of this study was to assess the influence of centrifugation on salivary sodium and potassium concentrations measured with pocket meters. Participants and methods: Samples of whole saliva were self-collected from 46 healthy volunteers and kept at -20°C before the analysis. Concentrations of sodium and potassium were measured using ISE handheld meters LAQUAtwin Na-11 and LAQUAtwin K-11 (Horiba UK Limited, Northampton, UK) in 91 saliva samples prior to their centrifugation and in post- centrifuged salivary supernatant. Samples were centrifuged at 1000 g for 5 minutes. In a total of 38 samples, measurements were performed in duplicate. Results: Intraclass correlation coefficient showed excellent reproducibility of data both in uncentrifuged and centrifuged samples. Concentration of measured electrolytes was higher in uncentrifuged saliva with p<0.001, and effect size of r=-0.855 and r=-0.487 for sodium and potassium respectively. Conclusion: When measurements are performed outside of the laboratories using portable devices and fresh samples, it is convenient to avoid centrifugation. However, the results of this study suggest that the concentrations of sodium and potassium in whole saliva significantly differ between centrifuged and uncentrifuged samples.
