Sažetak
Our previously published results suggest that maternal diet during gestation influences the subcutaneous and visceral white adipose tissue morphology in the offspring. Objective of this study was to further investigate the combined effects of maternal and offspring diet on morphology of the perigonadal white adipose tissue in rats. Materials & Methods:Ten female nine weeks old Sprague Dawley rats were randomly divided in two groups based on their diet, CD (control diet) and HFD (high-fat diet, food rich in saturated fatty acids) during five weeks. Thereafter, the females were mated with genetically similar males. Offspring animals from each group were further randomized into four subgroups based on their mother's and their own postnatal diet type: CD-CD, CD-HFD, HFD-CD and HFD-HFD. Harvested perigonadal (i.e. scrotal and periovarian) adipose tissue samples were fixed in 4% para-formaldehyde, paraffin- embedded, cut in 6 microns thick slices and stained with hematoxylin-eosin dyes. Histomorphometry and adipocyte quantification was performed in Cell Profiler. Results: Preliminary results show significant morphological differences between the CD-CD and CD-HFD, as well as between the CD-CD and HFD-CD rats in terms of hypertrophy (p = 0.016 and p = 0.038, respectively). Median areas of adipocytes in the CD-HFD and HFD-CD groups were 1918.16 μm² and 1623.46 μm², respectively, which indicates hypertrophy relative to the CD-CD group (1224.59 μm²). In male rats, adipocytes of the HFD-HFD group have a significantly smaller surface area compared to the CD-CD animals (p = 0.026), CD- HFD (p = 0.028) and HFD-CD (p = 0.013). The HFD- HFD male rats have adipocyte median surface areas of 1146.13 μm², the CD-CD group 2127.47 μm², the CD-HFD group 2007.68 μm² and the HFD-CD 19336.53 μm². Conclusion: Our results suggest that a change in postnatal diet relative to the gestational (maternal) diet leads to periovarian adipocyte hypertrophy in female offspring. In contrast to that, scrotal adipose tissue in male offpsirng was not significantly influenced by a postnatal change in the diet. Nonetheless, a combination of maternal and postnatal high-fat diet was significantly associated with hypotrophy of the scrotal adipocytes.
