Sažetak
Malignancies from the group of head and neck squamous carcinoma appearing in the larynx are considered devastating. Despite the fact that such cases represent 2-5% of all malignancies worldwide, they account for roughly 200, 000 deaths per year. The majority of larynx cancers, about 85-90%, are squamous cell carcinomas arising from the covering of the vocal cords. The complete elucidation of malignant disease pathogenesis, including the laryngeal squamous cell carcinoma (LSCC), remain elusive, especially when it comes to early loss of tissue homeostasis. Growing body of evidence connect circadian deregulation with cancer development and tumor progression, however, a clear association between circadian clock genes and LSCC is still lacking. Indeed, circadian rhythm deregulation seems to be an obvious element of malignant diseases as this rhythm is central in physical, mental, and behavioural changes that observe a daily cycle, including sleep and activity, appetite, hormone levels, metabolism, and gene expression. Keeping in view the importance of disrupted circadian rhythm in LSCC, we therefore, examined malignant and healthy adjacent tissues of patients suffering from LSCC to establish a correlation between the expression of the circadian clock genes (PER1, PER2, PER3, CLOCK, CRY1, CRY2, BMAL1, NPAS2, CK1ϵ, and TIM). Using real time quantitative RT PCR technique we evaluated whether the expression of circadian clock genes is altered in cancerous tissues among LSCC patients. Our results showed downregulation of PER1, PER3, CRY1, CRY2, BMAL1 and CLOCK. Among these genes PER1 and PER3 have statistically significant downregulation (p<0, 05), and the strongest downregulation has been seen for PER3. These results suggested a possible association between deregulation of circadian clock gene expressions, especially PER3 and LNSCC.
