Sažetak
Obesity is associated with excess body weight, cardiovascular disease, type 2 diabetes mellitus, hypertension and non-alcoholic fatty liver disease, all of which are connected with oxidative stress. Oxidative stress is a known factor for genomic instability that also impacts cancer development. Although conservative weight loss regimens demonstrated beneficiary influence on reduction of DNA damage, little is known whether a similar effect can be achieved with a more extensive weight loss using more restrictive calorie reduction diets, so predictions for human population are mostly based on in vitro and animal model studies. Little is known whether this type of diet would also show differences in biochemical parameters and also improve the whole health status of individuals. The aim of this preliminary study that included 10 obese volunteers with a body mass index of ≥ 35 kg/m2 was to examine the impact of a 3-week calorie reduction diet of only 500 kcal in strict hospital regime on genomic stability, oxidative stress levels and biochemical markers before and after the diet. Individuals had no known comorbidity and did not use medical therapy. Genomic stability was assessed using cytochalasin B-blocked micronucleus assay, alkaline comet assay and modified alkaline comet assay for oxidative DNA damage measurement. Biochemical parameters: glucose, triglycerides, total cholesterol, HDL, LDL and CRP were measured by an auto-analyser Beckman Coulter AU 480. Fresh blood samples were taken before and after the diet. The results demonstrated a reduction in body weight, oxidative damage, apoptosis and DNA damage in both alkaline and micronucleus assay, together with fasting glucose, triglycerides, total cholesterol, LDL and CRP reduction ; while HDL increased. The results indicated a beneficiary effect of this strict calorie reduction diet and should be repeated on a higher number of participants, with more biochemical parameters included together with the whole year diet questionnaire analysis.
