Type 1 diabetes (T1D) is characterized by autoimmune destruction of pancreatic insulin producing beta cells with the unknown cause, so any further information regarding the disease onset or development is beneficial. Glycosylation, as one of the most abundant co- and post- translational modification of proteins, is prone to changes in different diseases and thus considered as a potential biomarker of a disease onset or progress [1, 2]. In this study, we have tested the existence of age and gender dependent changes of plasma N-glycome in 1057 children and adolescents diagnosed with T1D. Plasma samples were collected through the Danish Registry of Childhood and Adolescent Diabetes (DanDiabKids). N-glycans were enzymatically released from plasma proteins using PNGase F, fluorescently labelled and then profiled using hydrophilic interaction ultra-performance liquid chromatography with fluorescence detection. Both age and gender dependent changes in N-glycome were found. In both genders, before puberty, a significant increase in low branched, a- and mono- galactosylated and core fucosylated glycans with age was observed. Also, a significant decrease in highly branched, di-, tri- and tetra- sialylated, and tri- and tetra- galactosylated glycans with age was observed. After the puberty, in female subjects, the majority of the aforementioned glycans changed the direction of their behavior with aging, while in male subjects only a decrease in tetrasialylated glycans correlated with age. These changes should be considered when conducting studies on potential glycosylation based biomarkers in children and adolescents with T1D.
