Sažetak
Transferrin (Tf) is a highly conserved plasma glycoprotein, known for its role in iron transport. Its synthesis occurs mainly in the liver. Tf is structured as a single polypeptide chain arranged in two independent metal ion- binding globular domains, each having one glycosylation site with mostly a complex biantennary sialylated N-glycan attached. It is known that variation of transferrin glycosylation has significant clinical implications in diseases like cancer, rheumatoid arthritis, galactosemia, in haemochromatosis patients and patients with heavy alcohol consumption. The aim of this study was to investigate whether normal physiologic changes during menstrual cycle affect Tf glycosylation patterns. The participants were healthy females with regular menstrual cycle, non- smoking and non-drinking history and not using oral hormonal contraceptives. Once a week, during period of 12 weeks, 5 mL of peripheral blood was drawn and plasma was separated from each female. The flow through fraction collected after immunoglobulin G depletion of plasma samples was used for Tf isolation by immunoaffinity chromatography on a recently developed CIM® anti-transferrin monolithic 96-well plate (CIM- @Tf 96-well plate). Tf N glycans were released by PNGase F, labeled with 2 aminobenzamide and analyzed by hydrophilic interaction ultra- performance liquid chromatography. Using this approach, we performed a longitudinal intrapersonal comparison of Tf glycosylation profile for different stages of the menstrual cycle.
