Quantitative evaluation of immunoglobulins in periapical cysts by radial immunodiffusion
The presence of immunoglobulins IgG, IgA and IgM was studied in human dental periapical cysts. Periapical cyst biopsy was performed in 16 subjects to assess their immune response. Sample tissues were homogenized in 0.5 mL PBS buffer and centrifuged. In the supernatant total proteins and immunoglobulin were determined. Total proteins were measured by Lowry's method. Immunoglobulins were detected by radial immunodiffusion method on LC plates. Valid data on the amounts of IgG, IgA and IgM were obtained on the basis of the measured concentrations of immunoglobulin in each periapical cyst biopsy and their correlation with total protein concentration, expressed as milligrams of IgG, IgA and IgM per gram of total protein. The mean values of total proteins found in periapical cyst samples were 3.12 g/L; IgG, IgA and IgM found in periapical cyst samples were in different amounts. IgG and IgA were detected in 100% and IgM in 33% of samples. Mean values of IgG, IgA and IgM in periapical cysts were 159.4, 25.6 and 1.8 mg/g of total protein respectively. The values of IgG, IgA and IgM found in periapical cysts suggested their possible local synthesis and indicated that non-specific inflammatory reactions may have occurred concurrently with specific immunological reactions.
