In vitro propagation of Alyssum samoborense horvat
A rapid clonal propagation for Alyssum montanum L. subsp. Pluscanescens (Brassicaceae) has been developed. A. montanum L. subsp. pluscanescens is an old autochthonous species inhabiting the coasts of former Pannonian sea, i.e. continental part of south-eastern and eastern Europe. Through the past, number of specimens decreased due to mountain elevation, soil erosion and climatic changes. Shoots originated from aseptically germinated seeds were used for culture initiation. Developed shoots were multiplicated on media containing full or half concentration of MS salts with 2.9 mM gibberellic acid (2.9 mM) and 6-benzylaminopurine (BA ; 0.2, 0.5 and 1 mM) or without BA. The highest multiplication rate was noticed on full strength MS medium enriched with 1 mM BA (2.85 shoots per explant). Before transferring to rooting medium, shoots were elongated on MS medium containing 0.2 mM BA and 14.5 mM gibberellic acid. After 4 weeks, elongated shoots were rooted on MS media containing different concentrations of auxins indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and a-naphtaleneacetic acid (NAA). The best rooting was achieved on media containing IAA (5.7 mM) and IBA (2.5 mM).
