Role of the small GTPases in the remodeling of the intracellular compartments during MCMV infection

Introduction: Alteration in cell surface composition is crucial mechanism for cells to adapt to environmental changes. Accordingly, it is expected for viruses to alter the intracellular trafficking of cell surface proteins in order to avoid immunological response of the host and to ensure intracellular environment suitable for viral assembly and egress. Murine cytomegalovirus (MCMV) reorganizes endosomal system of infected cell resulting in altered trafficking of many surface proteins. Considering that small GTPases are responsible for physiological function of endosomal system we wanted to test if expression and function of small GTPases from Arf family is altered during MCMV infection. Aims: The aim of this study was to determine the role of small GTPases from Arf family, especially Arf6 GTPase, in the reorganization of endosomal system and in alterations of intracellular trafficking during the MCMV infection. Materials and methods: All experiments were done on Balb3T3 murine fibroblasts. Cells were infected with 95.15 Δm138 MCMV and expression of Arf proteins and their effector was performed 0, 6, 16 and 30 hours post infection (hpi) by Western blot and by confocal microscopy. Expression of IE-1 viral protein was used as a control of infection. We used siRNA methodology for silencing the Arf6 protein. Results: Expression of Arf family member proteins (Arf1, Arf3, Arf4 and Arf5) is increased during the MCMV infection as well as membrane recruitment of their active forms (Arf6). Their effectors, like Big2 and EPI64 are redistributed and their expression is also elevated. Membrane recruitment of Rab35 and his effectors, as well as downstream Rab proteins is decreased. Conclusions: These results point to overactivation of Arf proteins during the MCMV infection, especially Arf6, which may be the cause of reorganization of endosomal system during the MCMV infection. Understanding these processes may help the development of therapeutic strategies.
