Morphometric parameters and antioxidative response of drought-exposed wheat seedlings

Due to the climate changes, drought become major abiotic stress factor that has negative impact on plant growth and production. Drought reduces the wheat production worldwide effecting yield and grain quality. From the physiological point of view drought causes osmotic stress resulting in decline of cell turgidity and dehydration which can be during the long-term exposure fatal for plant survival. Development stage of the plant such as germination and early developmental stage are particularly susceptible to water deficit which is manifested in growth and germination inhibition. As a consequence of drought application, plants produces a huge amounts of reactive oxygen species (ROS), a highly reactive molecules which can disturb the natural balance in the cell. In response to water scarcity, plants have developed some defensive mechanisms, including enzymatic and nonenzymatic systems as most common. Enzymatic defending system includes a large number of different enzymes where the most important antioxidative enzymes are non-specific peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX). The aim of this study will be to determinate the influence of drought in winter wheat genotype „Vulkan“ tracking the germination rate, shoot length, fresh weight (FW) and dry weight (DW). Study will be carried out during 7 days under controlled conditions using 10% polyethylene glycol-6000 (PEG) solution which represents the moisture deficit stress inducing media. We will also explore the response of young seedlings to PEG-6000 induced drought by measuring POD, CAT, and APX activity. This research will reveal the potential of the genotype under drought exposure in early developmental stage.
