A Review of Biological Activity of Compounds With Oxadiazole Core
Oxadiazole core is a heteroaromatic five membered ring system composed of one oxygen and two nitrogen atoms. It is a vital part of numerous biologically active compounds and is of great interest to medicinal chemistry in pharmaceutical drug development. Based on the position of oxygen and nitrogen atoms, oxadiazoles are generally classified in four groups – 1, 2, 3-oxadiazole, 1, 2, 5 oxadiazole, 1, 2, 4 oxadiazole and 1, 3, 4 oxadiazole [1, 2]. Among them, 1, 3, 4-oxadiazoles are especially interesting due to their versatile and useful biological properties, such as antibacterial, antiviral, antifungal, anticancer, anti- inflammatory, anti- hypertensive and antidiabetic properties. 1, 2, 3-oxadiazole core has also drawn attention in bioactivity testing and is already present in pharmaceutical drugs, such as Molsidomine, a long lasting vasodilatator, and Mesocarb, a long lasting psychostimulant [3]. Other compounds with 1, 2, 3-oxadiazole core have been tested for antibacterial, anti-inflammatory and antipyretic activity [4]. Studies have shown that 1, 2, 5-oxadiazoles also possess remarkable bioactivities, for instance antimicrobial, antiparasitic, anticancer and vasorelaxant activity. Research in the field of medicinal chemistry have produced novel compounds with furoxan or benzofuroxan moieties combined with classical drug structures in a single molecule. Thus, novel antiulcer drugs, calcium channel modulators and vasodilators were developed [5]. The aim of this work was to collect and analyze available literature on oxadiazole derivatives for their numerous beneficial pharmacological activities.
