Evaluation of Synergistic Antibacterial Efficiency of Tea Tree and Lavender Essential Oils Against Clinical Isolates of Staphylococcus aureus
Staphylococcus aureus is potentially pathogenic bacteria found in nasal membranes, skin, hair follicles, and perineum of warm-blooded animals. It may be a cause of a wide range of infections and intoxications. Essential oils (EOs) are complex mixtures of a wide variety of components and have been recognized for their antimicrobial properties. Antibacterial effect of different combinations of Tea tree (Melaleuca alternifolia) and Lavender (Lavandula angustifolia) essential oils against clinical isolates of S. aureus were evaluated using checkerboard method. Clinical isolates of S. aureus (30 strains) were collected during 2018 at the Clinical Hospital Centre in Rijeka. Bacteria were identified by standard biochemical tests and were stored at -80°C in glycerol broth. The MIC of the individual EO against staphylococci were determined by microtitre plate-based antibacterial assay. Then, the fractional inhibitory concentration index (FICi) was determined by checkerboard titration method. Results showed that MIC value for Tea tree EO was 12, 8 mg/mL, and for Lavender EO 25, 6 mg/mL, against most S. aureus isolates, including methicillin resistant S. aureus (MRSA). strains FIC and FICi values were calculated, based on the MIC values in order to express the antimicrobial effect of EOs in different combinations. All 60 possible concentration combinations were examined. The results of interaction types tests between the EOs showed synergistic effect (when FICi ≤ 0.5) in 17 combination of Tea tree and Lavender EO. Concentrations of EO in synergy were much lower than their individual MIC values. Tea tree and Lavender EOs combination exhibited synergistic antibacterial effect against clinical isolates of S. aureus, regardless of their antibiotic susceptibility profile, and may be used as potent natural antibacterial agent.
