Sažetak
There is increasing prevalence of obesity and overweight worldwide. Serotonin (5HT) plays a role in energy homeostasis and glucose metabolism, and is therefore important target in obesity. The Wistar-Zagreb 5HT rat model was developed by selective breeding toward extremes of platelet serotonin parameters. It consists of two sublines of rats with constitutionally altered serotonin homeostasis (high-5HT and low-5HT subline). High- 5HT animals have lifelong increased body weight and elevated adiposity as compared to low-5HT rats. To better understand the role of 5HT in pathogenesis of obesity and related metabolic conditions, here we examined functional and molecular response of Wistar-Zagreb 5HT sublines to specific metabolic challenge. Male (2 months old) and female (5 months old) rats from both sublines were fed high-fat diet (45% kcal from fat) for 11 and 8 weeks, respectively. During this time, we monitored weight gain of animals and assessed their glucose homeostasis by intraperitoneal glucose tolerance test (ipGTT). In both males and females, the low 5HT rats, as compared to high 5HT rats, displayed more pronounced weight gain and increase of plasma glucose levels in response to high-fat diet. Further, high-fat diet induced a clear increase of glucose levels in ipGTT in male rats of low- 5HT, but not high-5HT, subline. White adipose tissue and hypothalamic samples were obtained from all animals at the end of the high fat diet intake period and will be analysed for expression of regulatory adipokines (leptin, adiponectin) and several other genes involved in metabolic homeostasis and energy balance.
