BIOACTIVE COMPONENTS IN LEAF AND FLOWER OF BLACKTHORN: COMPARISION OF DIFFERENT EXTRACTION METHODS
Blackthorn (Prunus spinosa L.) is a perennial deciduous plant that grows all over Europe in the ecotone between forested areas and grasslands. Dried flowers and leaf of blackthorn contain high concentrations of bioactive components, the most prominent phenolic compounds such as flavonol glycosides, procyanidins and derivatives of hydroxycinnamic acid. Almost all parts of blackthorn find their application in medicine and food technology.1 A first part of our research involves the isolation of bioactive compounds from dried flowers and leaf of blackthorn using two methods - classical extraction (refluxing) and Soxhlet extraction in 70% ethanol or methanol. The obtained extracts is purified applying preparative thin layer chromatography chromatography (PTLC) and follows characterization of main components using FTIR - spectroscopy. In recorded spectra, absorption bands are attributed to the hydroxy, ester and carbonyl groups characteristic of the phenolic compounds, and in particular the derivatives of hydroxycinnamic acid such as chlorogenic and p- coumaric acid, which are present in leaf and flower of blackthorn. Results of investigation indicate the different content of bioactive components, depending on the solvent used for extraction. Increased content of phenolic compounds was obtained while using binary polar solvents (70% ethanol). Total phenols, flavonols and hydroxycinnamic acids in all extracts were determined using spectrofotometric method, while quantification of extract was carried out using high performance liquid chromatography.
