Influence of repetitive mild traumatic brain injury on some parameters of inflammation and stress in the mouse spinal cord

Aims: Amyotrophic lateral sclerosis (ALS) is a fatal progressive degeneration of brain and spinal cord motor neurons. Etiology of sporadic ALS is still largely unknown but epidemiological data and the results of some clinical studies suggest that one of its possible underlying causes is the exposure to repetitive mild traumatic brain injury (rmTBI). The aim of this research was to investigate the effects of rmTBI on the expression of some markers of cellular inflammation and stress in the mouse spinal cord. Methods: Adult, slightly anesthetized male C57BL/6 mice were subjected to rmTBI using the weight drop method (Kane et al. 2012). Brain trauma procedures were performed twice daily, 6 hours apart, during 5 consecutive days. Anesthetized but not traumatized mice were used as the control group. Mice were sacrificed 1, 3 or 7 days after the last brain trauma. Their spinal cords were dissected and prepared for the western blotting analyses. Results: In the spinal cords of injured mice increased expression of cyclooxygenase-2 was detected on the first day after the last head impact. Protein levels of inducible nitric oxide synthase and heat shock protein-70 were slightly elevated also on the first day, while the expression of nuclear factor kappa B was mildly increased on the third day following trauma procedures. Conclusions: Our preliminary data suggest that rmTBI in mice induces some inflammatory changes in the spinal cord during the first week after the last brain trauma.
