Somatic embryogenesis in callus culture of alfalfa

Three alfalfa (Medicago sativa L.) cultivars - Osjecka 66, Slavonka and Szarvasi-4 were tested in vitro for their capacity to prodoce callus and somatic embryos and embryo conversion to plantlets. Four medium protocols with three-step or four-step sequency medium were used in this study. Segments petiole and leaf young seedlings were the source of explants. All tested cultiovars produced callus on all medium protocols, and showed the ability to produce somatic embryos. The best embryogenic capacity and conversion to plantlets were observed in the cv. Osjecka 66, while the cvs. Slavonka and Szarvasi-4 showed somewhat lower embryogenesis response. Regenerated plantlets had normal morphological characteristics.
