Characterization and identification of cytosolic biomolecules that bind Cd, Zn and Mo in the gills of Vardar chub (Squalius vardarensis Karaman)
The early effects of metal contamination in freshwater ecosystem can be seen at the molecular and cellular level of organisms exposed even to low metal concentrations and thus they can serve as “early warning” indicators. Accordingly, the aim of this study was to obtain the information on Cd, Zn and Mo distributions among various cytosolic biomolecules in the gills of Vardar chub (Squalius vardarensis Karaman) exposed to metals in mining and agriculturally contaminated Macedonian rivers. To achieve this goal, cytosolic fractionation, before and after heat-treatment, was performed by size exclusion high performance liquid chromatography, metal measurements by high resolution inductively coupled plasma mass spectrometry, and biomolecule identification by two techniques of mass spectrometry (MALDI-TOF and liquid chromatography-mass spectrometry, LC-MS). Cadmium was eluted within two peaks with maxima at 90 and 20 kDa. The first peak contained heat-sensitive proteins, based on its almost complete disappearance after the heat treatment. The second peak was confirmed to contain heat-stable proteins, most probably metallothioneins (MTs), based on the same elution time as MT standard. Zinc was eluted within one peak with maximum at 90 kDa, which almost completely disappeared after the heat-treatment, confirming the heat-sensitive nature of comprised biomolecules. Association of Zn with heat-stable MTs was not observed. Cytosolic Mo was eluted within single peak with maximum at 7 kDa, which contained heat-stable biomolecules, and which was confirmed to have molecular mass of 3.3 kDa, according to MALDI-TOF analysis. Heat-stable biomolecules involved in binding cytosolic metals Cd and Mo were further identified by LC-MS.
