Gel Chromatrography of the Lipopolysaccharide from Salmonella Typhi on the Sepharose Gel
Samples of lipopolysaccharide from Salmonella typhi (strain O-901), obtained by disintegration of cell walls with lysozyme and by rinsing of whole bacteria with concentrated (1 M) solution of sodium chloride, were analysed. They were separated on a column of Sepharose 4B gel. Detection of different components in gel-chromatography fractions was carried out by UV absorptiometry, colorimetrically with phenol-sulphuric acid reagent, and by rmmunochemical methods. Chemical characterization of the samples and of characteristic fractions was made by the determination of total hexoses, rhamnose, and proteins. The lipopolysaccharide, obtained by the lysozyme method, was found to consist of a mixture of fractions of polysaccharide and proteinic character, with a wide range of molecular weights. The sample obtained by the rinsing of bacteria showed, in contrast, two distinctly separated components: one, with a high molecular weight, consisted of a pure lipopolysaccharide, while the other was most likely a complex of proteins and polysaccharides.
