Although the cancer therapies have improved significantly in recent years, the clinical use of chemotherapeutics is often limited due to chemoresistance and/or to undesirable toxic effects caused by non-specific cell killing at an effective dose of drugs. The modified nucleobases are subjects of numerous research because of their potential antitumor and antiviral activity. The aim of this study is to determine whether N-9 sulfonylurea derivatives of purine nucleobases differentiallly influence the growth of tumor cells growing in the two-dimensional and three-dimensional cell cultures. Inhibitory potential of the newly synthesized N-9-sulfonylpurine derivatives on human cell lines cultured in 2D (HeLa, Caco-2, NCI-H358, K562, Raji, HUT 78, MOLT-4 and CCRF-CEM, MDCK1) and 3D cell culture (HeLa, Caco-2, NCI-H358) was determined by colorimetric MTT test. Changes in cell cycle of treated cells and control nontreated cells were determined by flow cytometry. Obtained results show that leukemia and lymphoma cells are more sensitive on treatment with respect to carcinoma cell lines. Cells grown in 3D culture showed stronger resistance to tested compounds in comparison to 2D cell culture. Tested N-9-sulfonylpurine derivative R-194 significantly changed the proportion of Raji cells in specific phases of the cell cycle compared to control cells.
