Functional ES-derived neuronal networks: a comparison with dissociated hippocampal neuronal networks
Embryonic stem (ES) cells provide a flexible and unlimited source for the derivation of various neuron types. One of the key attributes of neurons is the ability to establish neuronal networks. We tested whether ES derived neurons are capable of generating functional networks and whether such networks generated in vitro are capable of processing information. Intracellular recordings confirmed that ES- derived neurons spontaneously fire action potentials (APs) triggered by synaptic potentials. Extracellular recording with planar multielectrode arrays (MEAs) demonstrated bursts of spontaneous APs propagating throughout the entire network. The statistical properties of APs within such networks were remarkably similar to those of adult neurons. ES cell derived neuronal networks could discriminate between stimuli of different intensity at a single trial level, a key feature required for efficient information processing.
