Identifying Pex21p as a protein that specifically interacts with yeast seryl-tRNA synthetase
Saccharomyces cerevisiae Pex21p is a peroxin involved in the peroxisome assembly (Purdue, P.E., Yang, X. and Lazarow, P.B. (1998) J. Cell Biol. 143, 1859-1869.) This protein also specifically interacts with yeast seryl-tRNA synthetase (SerRS), an essential enzyme required for serylation of tRNASer. SerRS/Pex21p protein interaction was identified by screening yeast cDNA library with SerRS as a bait in the two-hybrid system. The complex formation between these two proteins was also confirmed in vitro. Since eukaryotic aminoacyl-tRNA synthetases (aaRSs) are known to participate in assembles with other synthetases and non-synthetase proteins, we propose that the interaction with Pex21p may reflect another function of the peroxine, potentially related to the specificity of serylation.
