Mechanical and in-vitro performance of porous hydroxyapatite/poly(lactic acid)/polycaprolactone composite scaffolds
Hydrothermal transformation of aragonitic cuttlefish bone into hydroxyapatite (HAp) was performed while retaining the cuttlebone architecture. Composite structures were obtained using vacuum impregnation technique with different PCL/PLA ratios. Final three-component systems were characterized by X-ray diffraction, FTIR spectroscopy, TGA and SEM. Influence of different polymer ratio on mechanical properties was investigated by compressive test, while cytotoxicity of materials was evaluated by MTS assay. Results X-ray diffraction, FTIR spectroscopy and SEM confirmed thermal conversion of aragonite to HAp maintaining original cuttlefish bone microstructure. Compressive test showed that the PCL/PLA-coated HAp scaffold resulted in a material with improved mechanical properties with interconnected porous structure indispensable for tissue growth and vascularization. Variation of PCL-PLA ratio has resulted in alteration of scaffolds elasticity. Additionally, prepared PCL/PLA-HAp systems are non-cytotoxic.
