Ultrasound and microbubble assisted siRNA delivery in breast cancer cells
RNAi-based gene therapy for cancer treatment has not shown significant clinical impact due to poor siRNA delivery to the target site. In this work we used the ultrasound and microbubble-assisted siRNA delivery in breast cancer cells (murine mammary gland carcinoma constitutively expressing luciferase). In vitro studies were performed to confirm down-regulation of the luciferase reporter gene expression by our delivery system. With ultrasound and microbubble-assisted delivery we can target a specific area with elevated concentrations of therapeutic nucleic acids bound to cationic microbubbles, anticancer drugs or a combination of both with minimal invasiveness to the treated tissue. We have shown that cationic microbubbles can complex siRNA molecules and efficiently transfect the cells during an ultrasound cell treatment. Our results offer a proof of concept for ultrasound and microbubble-assisted delivery of gene-targeted siRNA. We also produced cationic microbubbles that were able to carry both nucleic acids and a chemotherapy drug, paclitaxel. Such microbubbles have also been subjected to in vivo testing.
