Pyrrolocytosine-pyrene hybrids as new chemical receptors for the molecular recognition of nucleic acids

Small fluorescent molecules targeting DNA/RNA have attracted significant scientific interest due to the medicinal, biochemical and biological implications of such molecular recognition events. According to the previous results, we designed and synthesized new type of molecular probes, containing pyrene as fluorophore, as well as DNA-intercalator unit and fluorescent pyrrolocytosine (Pyrr-C), which should by Watson-Crick base-pairing capacity recognise guanine. Three novel Pyrr-Cpyrene hybrids 3 and 4, as well as bis(pyrenyl) hybrid 5 were synthesized. The spectrophotometric properties of compounds 3-5 and their interactions with ds-DNA and ds-RNA were studied by UV/Vis, fluorescence and CD spectroscopy. Their bioactivity and cellular uptake will be tested on a panel of human tumour cell lines.
