Fluorophore one-atom-controlled recognition of DNA/RNA secondary structures
Cyanine dyes are commonly used as molecular probes for nucleic acids and proteins due to well known fluorescence “switch-on” upon binding to target, while nonfluorescent in free form. Here we present two novel cyanine-amino acid conjugates differing only in a halogen substituent, prepared by Cu-catalyzed azide- alkyne cycloaddition. Remarkable difference between fluorine- and chlorine- analogue interactions with polynucleotides was documented by circular dichroism spectroscopy and additionally analysed by molecular modeling simulations. Biological experiments in the cells showed that studied conjugates are not localised in nucleus as parent analogous cyanine dye (TO), but target mitochondria.
