Characterization of the ribosomal RNA operon rrnF from Streptomyces rimosus
Restriction sites for BamH1 endonuclease do not exist in S.rimosus rRNA operons. Digestion of S.rimosus genomic DNA with BamH1 leaves sic operons (A-F) on six DNA fragments. The smallest fragment is 8.8 kb long and contains rRNA operon named rrnaF..We cloned this operon into pBR322 and characterisation of its primary structure is in progress. Over two thousand nucleotides are already sequenced partially covering16S and 23S rRNA genes and whole 5S rRNA gene (116 nucleotides). Genes are organized in the order 16S-23S-5S. 5S rRNA gene is followed by two long inverted repeats, structure characteristic for rho independent terminator. tRNA genes are not located at the 3'end of rrnaF operon. Two very conserved Pst1 sites 4.5 kb apart, foud in all six rRNA operons, are located around position 465 in 16S RNA gene and position 85 in 5S RNA gene, which means that in all rRNA operons spacers between genes are short and of the similar size.
