Assesment of adjuvants - the effect of peptidoglycan monomer and adamantyltripeptides on humoral immune response in mice

Peptidoglycan monomer*, GlcNAc-MurNAc-L-Ala-D-isoGLN-meso-DAP (wNH2)-D-Ala-D-Ala (PGM) originating from Brevibacterium divaricatum exhibits immunomodulating activity (1). Synthetic adamantyltripeptides, isomers of D,L-(adamant-2-yl)-glycyl-L-Ala-D-isoGln (AdTP1 and AdTP2) were also shown to posses immunomodulating properties (2). The purpose of the present work was to asses the effect of these compounds on humoral immune response in an experimental model suitable for comparison of adjuvant activity. BALB/c mice were immunized with ovalbumin (OVA). Control groups received OVA in saline (50 mg/mouse) and separate groups received respective tested compounds (100mg/mouse) in addition to OVA (also in saline) only in the first injection. Booster injections of OVA were administrated on days 14 or/and 28. Anti-OVA was assayed by ELISA, developed for this purpose, in the sera taken from mice 7 days after the booster injections. After the first booster, the sera of mice treated with PGM and AdTP2 exibited significantly higher anti-OVA levels (expressed as absorbance at 492 nm at different sera dilutions) than the control group or the group treated with AdTP1. After the second booster the levels of anti-OVA in groups treated with PGM and AdTP2 were also higher than in control group, but the difference was not statisticaly significant.
